The assessment combines a detailed semen analysis with hormonal profiling, infectious disease screening, blood group determination, haemoglobin genotype, and a full blood count. All testing is performed using validated laboratory methods, including microscopy and computer-assisted semen analysis, immunoassays for hormone and infectious disease markers, and standard haematological techniques.

Semen Analysis
- Sperm count: the number of sperm present in the sample, a key indicator of fertility potential
- Motility: the percentage of sperm that are moving, and how well they swim
- Morphology: the percentage of sperm with a normal shape, which affects their ability to fertilise an egg
- Volume: the total volume of the semen sample

Hormonal Profile
- Prolactin (PRL): a hormone that can affect sperm production when elevated
- Luteinising hormone (LH): stimulates testosterone production
- Oestradiol (E2): an oestrogen that, in men, can influence sperm production when imbalanced
- Testosterone: the primary male sex hormone, essential for sperm production and libido

Infectious Disease Screening
- HIV: human immunodeficiency virus
- Hepatitis B: hepatitis B virus surface antigen and core antibodies
- Hepatitis C: hepatitis C virus antibodies
- Syphilis: treponemal antibodies
- Rubella immunity (IgM/IgG): checks immunity to rubella, which is important for the safety of the mother and baby
- Varicella immunity: checks immunity to chickenpox, which can be harmful in pregnancy
- Chlamydia: a common sexually transmitted infection that can affect fertility
- Gonorrhoea: a sexually transmitted infection that can affect fertility

Blood Group & Type
- ABO: determines the ABO blood group
- Rh: determines the Rh factor (positive or negative), important for pregnancy management
